Med-Medicine - Pennsylvania-Pittsburgh - (23009354) The Division of Pulmonary, Allergy, Critical Care and Sleep Medicine is looking for a Senior Research Technician. The position requires performance of specialized techniques to understand various aspects of lung disease with focus on asthma and acute respiratory distress syndrome (ARDS). Experiments will involve:1) Processing of human samples from healthy subjects, asthma, ARDS, and sepsis patients for immune cell enrichment for immediate use and cryopreservation. The samples will include blood and lung lavage. Experiments will be performed with enriched cells.2) Implementation of a new technique of high dimensional flow cytometry for analysis of human and mouse immune cells.Job SummaryPerforms a variety of independent and complex laboratory work and experiments. Analyzes and interprets data, compiles results, and prepares reports. Maintains inventory, ordering, and logs. Writes manuscripts and grant proposals and prepares data visualizations.Essential Functions 1. Processing of human samples from lungs and peripheral blood by density gradient centrifugation and further processing for cryopreservation using specialized protocols. 2. Breeding of genetically modified mice and genotyping. 3. Setting up mouse models of lung diseases and analysis at the completion of the experiment. 4. Analysis of human and mouse samples by qRT-PCR, ELISA and related techniques.5. Acquisition of a new sophisticated skill-set of multiparameter, high dimensional flow cytometry for immune cell profiling. 6. Analysis of tissue by histology, immunological, and microscopic techniques. 7. Equipment and reagent maintenance and ordering of lab supplies. 8. Ensuring compliance with University and Federal Government rules for proper functioning of the lab. 9. Supervision of lower-level technicians, undergrad students, clinical (pulmonary) fellows, and other trainees in the laboratory. Development of training programs to facilitate the acquisition of laboratory skills by trainees and new hires.Physical Effort - Must be able to stand and/or sit for up to 8 hours.
